生产方法和用途
方法
8.9g 2-(N-甲基-N-(2-吡啶基)氨基)乙醇溶于60ml二甲基甲酰胺,在氮气保护下,分批加入0.7g 60%的氢化钠。当剧烈的反应减慢后,加入2.9g 4-氟苯甲醛,在80℃加热16h。冷却,加入400ml水。用3×250ml乙醚萃取,萃取液合并,用2×100ml食盐水洗,干燥,过滤,浓缩至干,得到化合物(Ⅰ)。3.2g化合物(Ⅰ)和1.1g 2,4-噻唑啉二酮溶于100ml甲苯,加入催化量的哌啶乙酸盐,回流2h。冷却,过滤,得到化合物(Ⅱ),熔点177-190℃。化合物(Ⅱ)溶于二氧六环,在10%Pd-C催化下,在室温和常压下氢化至氢气吸收停止。过滤,减压浓缩至干。剩余物用甲醇重结晶,得罗格利酮,熔点153~155℃。
用途
用于治疗糖尿病
噻唑烷二酮类胰岛素增敏剂。适用于非胰岛素依赖型(Ⅱ型)糖尿病的治疗。
